Baking question: sugar substitutions?

Hi, I was skimming through recipes that I would like to try making, mostly muffins, and was trying to think of ways to make them healthy.
Has anybody substituted splenda for sugar, or whole wheat flour for regular flour and have it go terribly wrong?
Do you have tips for cutting calories on baking - ex substituting less of one ingredient and adding another that tastes very similar?
Can you substitute any ingredient for butter or margarine and have it taste the same?
I'm a very novice baker and require any and all tips. Thanks!!
